General Description: An experienced peptide/protein chemist with an interest in the structure and function of G protein coupled receptors is sought. The position requires a PhD in Chemistry or Biochemistry with knowledge in peptide/protein synthesis and biosynthesis. In particular the candidate should have experience in molecular biology and protein expression. The project involves the biosynthesis of large fragments of a G protein receptor from yeast, the isotopic labeling of these fragments and the reconstitution of the protein for spectroscopic (CD, fluorescence, high resolution NMR)evaluation. The goal of the research is to determine the structure of domains of the receptor in the context of the entire receptor. Other Duties: The major duties will be construction of expression vectors for Ste2p the GPCR from yeast, transformation of cells with these vectors, expression of fragments of Ste2p directly or as fusion proteins, isolation and purification of the target fragments, reconstitution in membrane mimetic environments and assay of binding of the reconstituted materials.Salary:$32,000-34,000 per year Core Competencies/Qualifications: 1) Protein biosynthesis 2) Recombinant DNA experience 3) PhD 4) Peptide/protein purification 5) HPLC 6) CD 7) NMR Position will remain open until filled.
