Natively, American-issue Genesis hardware does not support S-video output.
You can't get it from the multi-out, and the X'eye doesn't have it either.
Eagle-eyed users have noticed an extra stamp-out circle on the back of the
X'eye, which is an indicator that some of the Japanese Wondermega variants
do suport S-video. If you're handy with a soldering iron, it's actually
relatively easy to tap the S-video signal from the video encoder chip
inside any Genesis.

The big reason for wanting S-video from the Genesis hardware is higher
color fidelity. Sega's composite output on the Genesis and the Saturn is
already really high quality, so using S-video isn't as much of a step up
as it is for other game consoles.
Oh yes, here's a link to diagram for S-video on later Genesis hardware:

The process for getting S-video on a Mark I Genesis is a little more
involved, but thankfully it's really well understood. Incidentally, it's
the same mod used for the Neo Geo home console:

-KKC, who should really put some of this useless knowledge to work some
day.
